Yes. You will have to do multiple water changes to remove it. I would not run the skimmer during this period. There's no reason to. It's only going to overflow.

This stuff definitely works. Woke up this morning I find that 85% it the cyano was gone. Gonna do the water change tonight. Should I do a second dose to get rid of the rest or wait a few days?

No. Follow the directions exactly. It will all be gone in a few days.
