So, Can I start my 90 gal glass, undrilled tank with only a HOB skimmer, HOB refugium, 100 lbs Fiji LR, a 4in DSB and 48in MH FL ix lighting at 400w?
 
I dont know why not. A sump would be ideal but you can make it without if you must.

Is it possible to do a sump with a tank that is not drilled? Is the sump better because of capacity?
 
There are hang on overflow boxes that work well if the proper size return pump is used.

Typically sumps allow for higher quality skimmers and the possibility of a refugium of larger volume underneath.
 
Yes Reefermadness you dont have to have a drilled tank as HOB overflows will work just as good. I had one in the first few years I had a tank ,before my 125. And yes they (sumps) provide more water volume which is always good. There is always other benefits as a place to put your equipement and a fuge if you so desire.
