When to do my first water change

Had my 25 gallon set up for 3 weeks with 25lbs of live rock. The ammonia has just gone down to almost nothing and i have 3 damsels and a cleaner shrimp inside now. When should i do my first water change and how many percent i should do becoz i will bring back those damsels to the fishstore and get a clown fish or an flame angel with an anemone.

Once nitrites hit zero you will be good to go. Most say a 50% change would be in order. I would holdoff on an anenome until your tank is a little more established

When the amm and nitrite are zero do a pretty good size change(15 to 20%) then get on a regular schedule of 10% or so a week. I would wait on the anemone until your tank settles in a little though. Give it at least a few months. Make sure you have enough lighting for whichever one you decide to get.

Im planning to buy a flame angel and a clown fish, then im going to take the damsels out of the tank. Should i do the water change before i put the angel and clown in or should i put them in then do the water change or does it matter?

I would say a flame angel is too large for a 25gallon tank. It hits 4" in length...I'd keep it in nothing under 30gallons...though 40+ would be 'ideal'.

Any amount of ammonia or nitrite is bad for fish...that's why cycling with fish is never recommended.

If you plan to swap fish, I'd quarantine the clown for 2-4 weeks. If you introduce a sick fish (for example, one with marine ich) to your display tank, you'll have to remove the fish to medicate it, and then run the display tank fallow (without fish) for 6 weeks to ensure no parasites are still alive.

However to directly answer your question, change the water first, then add the fish. Acclimation is stressful, and following it with a water change could push the fish over the edge.
