Question about glass diffuser

I just bought this glass diffuser, but no real instructions came with it.
am I supposed to fill the inside with water?

I have the exact same diffuser that I use in my 30G tank with pressurized CO2 and it does a great job. Don't worry about filling it with water, the water will get into it when you submerse it. As long as you have sufficient CO2 pressure pumping into it you should see a very fine mist of CO2 bubbles coming out of the top of it once the CO2 starts flowing.

Are you using pressurized CO2 or DIY?
 
I've never used DIY before so I leave it to the DIY experts to answer definitively, but my guess would be that it will take a while before you get enough gas pressure to force all of the air and water out of the tube and diffuser before you start seeing the CO2 mist coming out. I run mine on pressurized CO2 at two bubbles per second and it does an excellent job.
 
I'm currently using that diffuser on my 29G. I have it placed directly under the XP2 intake. Works great. But not sure if DIY will have enough pressure. It's worth the try though.
