If you can access the back of the tank when it is filled I don't see why not. Just cover all openings very well. As long as paint stays out of the tank you should be fine. You don't want to paint the inside of a filled tank though, I'm not sure if that's what you meant or not lol

IMO, painting is definitely better and long as you use water based paint (long as it is not oil based) it is so easy to remove....IME...
I painted mine before i had fish in it, but i used plastidip (found at homedepot). I used it because if i ever want to remove it all i have to do is peel it off. You can get different colors of it if you order it online. It took me 1.5 cans for a 55gal.
