I was handling my 40 gal aquarium with a buddy, and later when inspecting it I noticed in the top most corner what appeared to be a crack. Is this going to be an issue for me? Do I need to get this pannel replaced?

Hello, you can take a glass cutter & etch it deep across end(like a T)of crack to keep it from spidering. Some people do this on windshields. But remember 1gal of water weighs 8.34lbs & a 40gal holds 334lbs water. It might fail. Hope this helps you.
