Is my skirt tetra pregnant? If so what should I do?

Good news is they aren't pregnant, since they are egg-laying fish. It could possibly be holding eggs? Pm andy sager he has lots of knowledge on fish breeding

If it is holding eggs what should I do with it?

Theres nothing you can do if you look down on the fish you may be able to tell the sexes. The rounder ones can be females and the slimmer, more streamlined ones male. Maybe look up their breeding info and follow that?

Put her in a smaller tank and wait till they lay then move her back to the bigger tank?

Nah because they haven't been fertilised
