I added 3 new glassfish to my 55g to finish the school and want to identify their genders. I know the makes get bigger but is a black spot behind their gill also a sign? The 2 I had before are fully grown(as far as I'm aware) and the male also has this spot.

Males will have a blueish edge on their dorsal and anal fins, I've never heard of a black spot being a factor.

So I checked the young ones I have. All the ones with a black spot also had blue on the tips of their dorsal fin.

I just ran across something while looking for a possible reason that your fish are so big. It seems that males do develop a black tip on the swim bladder so thumbs up on the observation.
