Adding Prime to "aged" water

I've got into the routine of filling a water barrel 48 hours before I do a water change to allow it to degas and get up to temperature before doing the water change (I put a heater in there).

Even though the chlorine should bubble off by itself during that time, I still add Prime just in case there is any left, and also in case there is any chloramine in the water (our municipality says they don't use it but that might change without notice)

Now seeing as Prime neturalises chlorine, nitrates etc for 48 hours, when would the best time be to add Prime? When I fill the barrel and let it stand, or just before I do the water change? or doesn't it make any difference?

Is there a danger that if I add it too soon, it's effectiveness would be worn off by the time I do the water change?
 
I add 'safe'(dry form of prime) to my prep barrels when I fill them.

It works almost instantly on chlorine(I have test kit).
 
The effect on nitrogen is temporary but not the dechlorination. Add it whenever you like.
