im wondering if i could use multiple substrates in one big fish tank.

i dont mean like mix them together

like i line them up as in columns.or rows

one area for soil for plants that needs it. the other for play sands, white sands, and the rest the area for decor' sands.

does this work?

When doing wc you will mix them up so it will be tough

You can easily do a full bottom layer of organic potting soil with a cap of sand, commercial plant substrate, or small grained gravel. Some people add and area of different colored sand to make paths or areas for sand loving bottom dwellers but these applications will very quickly be messed up and intermixed mostly from fish or when your planting.
