On my five gallon tank one of the sides cracked I was wondering what's the best and simplest way to repair it.

The simplest way is not the best and the best is not the simplest. It really needs to be broken down totally, all the silicone taken off (new silicone does not seat properly with old) take the frames off, replace the panel, reseal, reinstall frame.

Do you mean take all 4 panels off or just one?

Honestly after all the work and cost for a replacement piece, it may be a lot easier to just get a new tank. You'd want to remove all the silicone off of the contact points of the panel that you are replacing
