You've misunderstood me. There is nothing in the tank, there is no waste. I want to know if removing the skimmate is required TO BREAK IN the skimmer. I know what a protein skimmer does and how it works.

A dead shrimp will show ammonia readings within 24 hours. If you have shrimp in there and your ammonia and nitrite are 0 then I would say toss the shrimp and you are good to go.
