The best thing you can do for it is put it in another tank all by itself. It's still in danger if you leave it in the tank. Can't help you with the guppy thing. Maybe the females are just larger. That's normal in some species of fish
Do you have a carbon filter cause that's why your treatment isn't working.for heat in a larger tank like that you should have two heaters rated for a tank half the size of yours instead of one big one
