You absolutely do not need bacteria in a bottle to cycle a tank. Biggest thing with a cycle is to bind lethal toxins and let nature take its course.

I can't really tell. I have had females with similar guarded positions, but only when dealing with males/ nursing wounds from males. If it's not external only or constipation I'm baffled... Have you tried feeding her a pea?
