When should i test i just did a 50% water change and want to know how long to wait before testing for PH and KH trying to get my Co2 level thanks

You'll need to wait at least a couple of hours, 24hrs if you want to be absolutely certain that there's no interferance from the water change.

ok and one more question someone probably covered this but i want to see how much Co2 is actually getting dissolved take tests with the light off or on?

To get a true picture of you CO2 you'll want to do several tests. One right before lights on (this should be the highest), some periodically during your photo period, one right after lights off (this should be the lowest). You may also want to test several spots within the tank as the CO2 concentrations may vary significantly depending on the current in the tank.
