Water testing

Does it matter where you get your water sample from? I always take a sample from the top of the tank. I have pretty good water filtration/movement but it's a tall tank. Would this change my results? Would love to hear your thoughts fish peeps.
 
It's recommended you insert the test tube upside down until the middle of the tank, then turn the test tube up to fill with water, remove and let excess water out of the test tube before testing. I wouldn't worry to much for a normal tank, though a tall tank would be different.
