After 12 hours, ammo went down to .50 from 4.0 and the nitrites and nitrates have not moved. Let it go another 12 hours and check it, or just dose back to 4ppm? I think its safe to assume that if I leave it, nothing will change lol.

You can drain the water, refill with dechlorinated water and add the Puffer and finish with a fish-in cycle. Just be diligent with daily water tests and water changes as needed for as long as they are needed. It's up to you and how much commitment you're willing to put into the tank with a fish in there.
