How long do you wait after a water change to test amo?

I had an ammonia spike. It was at 0.5 and so I vacuumed and changed about 40% of the water in the tank (55g). My question is how long do I let the tank alone before I test the ammonia again? How soon would be too soon for another large water change. I think the spike is due to over cleaning the canister filter and then adding fish.
 
I waited an hour and retested. It's between .25 and .5 now

I would have done a larger change but since you are still above .25 I would another 50% about 2 hours after the last one. Dont vacuum again so you arent disturbing the fish any more than nessary.

I just changed about 60% of the water and added my Prime. I'll do another test in about an hour once old and new water are mixed together well. This really is not cool. I've lost 4 fish.

I have an Eheim pro filter that I hate and I have an aquaclear coming on Thursday for extra filtration

Just retested and ammonia is down to .25. More water changes are in store tomorrow!
