It should work in seconds, but I'm not completely sure. I use Start Right by Jungle, and that works in seconds

You can put it in right away. I hold mine for 2-24hrs, because pH out of the tap is 8-8.2 and after 2hrs locks in at 7.8 (same pH in all my tanks).

Sure does. If you have a big tank and use a python for PWC, put the Prime into the tank (filter off) and then add water from tap.

Ahh nope no python.. Do it the old fashion way, i usually let it sit in the bucket for 20-30 min, just to burn off any extra chemicals not needed.
