Water chemistry?

When testing parameters, will testing water from bottom or top of tank have different readings?

What do you suggest?

It shouldn't matter if you have decent filtration.

Did an fish in cycle and was reading 0 ammonia, 0- nitrite, and 5-10 nitrate.
It's been about a week so was really surprised at the positive readings.
Thought it was bogus
