If my aquarium ammonia reading is not at 0, can I add Seachem prime directly to the water?
Thanks

While NOT the best way to handle the situation it can be done, because I have done it. But I would suggest doing a PWC as soon as you can. I've added Prime with no PWC when I did not have time to do a PWC & I wanted my fish to be safe until the next day when I could do a PWC.
