So I'm starting up a 50 long. I want to do the PFS for the bottom. So do I put the sand in first, or fill it with water first, or does it matter?
 
PFS is good because it's inert and doesn't effect pH.
You want to wash the sand out really really well to make sure all of the dust particles are out.
I washed my sand probably 30 times in total before putting it into my tank.
Then you just put the sand in and fill the tank.
You should put a bowl or plate or something on the sand to dump the water onto so it doesn't send your sand flying and put a big crater in the bottom.
